Sequencing Methods
Samples will be subjected to Edman chemistry by the appropriate
method as dictated by the way in which the sample was presented to
the Protein Chemistry Laboratory (Method 3.1 for liquid protein
samples; Method 3.5 for liquid peptide samples; Method PVDF 3.1 for
electroblotted protein samples). The details of all current methods
are available in our laboratory.
All samples are preceded by PTH amino acid standards for
calibration purposes. The ternary HPLC system and chromatographic
method separate the 19 standard PTH-amino acids, sequencing
by-products and several modified forms of PTH-cysteine to greater
than 98% resolution. Retention times of the standards are used for
peak identification. Customized reports are generated using the HP
Chemstation software giving chromatograms of each cycle, repetitive
yield graphs and histograms. A blank HPLC trace is included in all
experiments to evaluate background signal. Blank runs are included
in the raw data along with your sample.
